Rear Inner Tube size for stock 2006 klx250s tire?

Also be aware, adding rimlocks will throw your tire balance seriously out-of-whack for street riding; so either add two to each wheel, or order some spoke weights at the same time. I ended up with Motion Pro Litelocs and only needed a couple ounces of weights per wheel (vs. 4 or 5 for the heavier locks). I used wieghts from 'THE REUSABLE SPOKE WEIGHT STORE' on fleabay... good stuff.
As for 'How loose?", just run the nut up to the cap on the schrader valve. Every now and again, take a look at the valve and see if it is at an angle... that way you know when your tire is rotating on the rim and can correct it before losing the stem.

You asked about size in the title. If you need it, most tubes are multi size due to the obvious stretch in rubber. If you get one that will work with a fit for 4.00, 4.50, 110 or 120 in the labeling you will be good to go.
