Whats the difference-spokes and rims for a...
KDX220r front wheel and a KLX250s front wheel? They have different part numbers for the rim so is it larger/thicker spokes and holes in rim for the KDX220 wheel? They appear to be interchangeable as a whole unit with hub-spokes-rim and speedo drive?

KDX front 95+ is what you want. Same brake rotor mounting (but a different rotor), same axle, same odo drive. The rims are also the same size. . If they have different part numbers they are cleary different parts but I am here to tell you they are interchangable & work.
& by that I meant installing the complete KDX wheel to the KLX. I vener tried mixing & matching rim/spoke/hub combos. The brake rotor, odo drive, & axle are all interchangable though like I said.
Ok, good to know as I got me a used one as I read they would work on the KLX once a long time ago on this forum I think. This one has a bent rim so I may have to buy a rim. So I just need to buy one for the KDX (as opposed to the KLX) so as not to mix spokes and hole sizes etc...Can I use the brake rotor from the KDX on the KDX wheel on the KLX or do I need to swap the disc??
