I use a 15 tooth on my 331cc on road sometimes (SM Kit ). Nice tall gearing. Not so cool off road when I use trail set up. Too tall in first and second for gnarly climbs.
However having said all that, on a standard 250 you may battle a bit pulling at top speed in 6 gear. I doubt if you will rev out, except down hill. ( You may find you are faster in 5th. 6th becomes a cruising overdrive) It will make the whole on road experience more pleasant and a lot less 'buzzy'.

more tooths to the sprocket near engine (front sprocket) means longer gear ratios.
more tooths to the rear sprocket (near the wheel) means shorter gear ratios.
i wouldnt recomend a 15 front tooth for a klx 250.. It would be way longer than it sould be and very bad for dirt riding... the stock 14 sprocket is ok for tarmac use and less dirt use, imagine what will happen if you use 15.

It would make a marginally-slow bike even slower, clutch slip more to get moving and reduce the rpm a bit at higher cruise speeds. It wouldn't necessarily improve mileage *noticeably* at higher cruise speeds, given that the throttle may have to be held more open.
All said, it's a cheap thing to try out to see if it suits you....and I see you're in Nevada so the mileage may get a tad better if you keep the speed down
